Abstract
ObjectivesTo compare the safety and tolerability of valacyclovir (Valtrex) and acyclovir (Acyclovir) after two doses at two doses on two different days. Methods: A prospective randomized, crossover study of the safety and tolerability of valacyclovir and acyclovir after two doses on two different days compared with a placebo for 12 weeks. Patients received either valacyclovir, acyclovir, or both. Acyclovir was given once every 12 weeks for 12 weeks. At 12 weeks, patients were randomized to receive either valacyclovir, acyclovir, or both valacyclovir and acyclovir in a double-blind fashion. At 24 weeks, patients were randomized to receive either valacyclovir, acyclovir, or both valacyclovir and acyclovir in a double-blind fashion. Results: The safety rate of valacyclovir and acyclovir was significantly lower than the rate of acyclovir. In addition, the rate of side effects did not differ between valacyclovir and acyclovir. Conclusions: Although both valacyclovir and acyclovir had a lower rate of side effects, the rates of adverse events did not differ. Valacyclovir is generally well-tolerated, and both valacyclovir and acyclovir have a lower rate of adverse events. Patients should be aware that valacyclovir is not recommended for patients with kidney disease. The dose of valacyclovir and acyclovir should be given every 12 weeks. Acyclovir is generally well-tolerated, and both valacyclovir and acyclovir have a lower rate of adverse events than valacyclovir. Because these drugs are not approved for the treatment of cold sores or genital herpes, there is no evidence that valacyclovir is effective for cold sores.
